  • COLOSTRUM DESCRIPTION

    Colostrum is the thin, yellowish, milky fluid secreted by the mammary glands of mammalian mothers (including humans) during late pregnancy and the first 24 to 48 hours after birth before true milk appears.

    HUMAN COLOSTRUM

    Newborns have very immature digestive systems, and colostrum delivers its nutrients in a very concentrated low-volume form. It has a mild laxative effect, encouraging the passing of the baby's first stool, which is called meconium. This clears excess bilirubin, a waste-product of dead red blood cells, which is produced in large quantities at birth due to blood volume reduction from the infant's body and helps prevent jaundice. Colostrum is known to contain immune cells (as lymphocytes) and many antibodies such as IgA, IgG, and IgM. These are some of the components of the adaptive immune system. In preterm infants some IgA may be absorbed through the intestinal epithelium and enter the blood stream though there is very little uptake in full term babies. This is due to the early "closure" of the intestinal epithelium to large molecule uptake in humans unlike the case in cattle which continue to uptake immunoglobulin from milk shortly after birth. Other immune components of colostrum include the major components of the innate immune system, such as lactoferrin, lysozyme, lactoperoxidase, complement, and proline-rich polypeptides (PRP). A number of cytokines (small messenger peptides that control the functioning of the immune system) are found in colostrum as well, including interleukins, tumor necrosis factor, chemokines, and others. Colostrum also contains a number of growth factors, such as insulin-like growth factors I (IGF-1), and II, transforming growth factors alpha, beta 1 and beta 2, fibroblast growth factors, epidermal growth factor, granulocyte-macrophage-stimulating growth factor, platelet-derived growth factor, vascular endothelial growth factor, and colony-stimulating factor-1.

    Colostrum is very rich in proteins, vitamin A, and sodium chloride, but contains lower amounts of carbohydrates, lipids, and potassium than mature milk. The most pertinent bioactive components in colostrum are growth factors and antimicrobial factors. The antibodies in colostrum provide passive immunity, while growth factors stimulate the development of the gut. They are passed to the neonate and provide the first protection against pathogens.

    ANIMAL HUSBANDRY

    Colostrum is crucial for newborn farm animals. They receive no passive transfer of immunity via the placenta before birth, so any antibodies that they need have to be ingested (unless supplied by injection or other artificial means). The ingested antibodies are absorbed from the intestine of the neonate. The newborn animal must receive colostrum within 6 hours of being born for maximal absorption of colostral antibodies to occur. Recent studies indicate that colostrum should be fed to bovines within the first thirty minutes to maximize IgG absorption rates.

    Colostrum varies in quality and quantity. In the dairy industry, the quality of colostrum is measured as the amount of IgG (Immunoglobulin G) per liter. It is recommended that newborn calves receive at least 4 quarts (liters) of colostrum with each containing at least 50 grams of IgG/liter. Testing of colostral quality can be done by multitude of devices including colostrometer, optical refractometer or digital refractometer.

    Livestock breeders commonly bank colostrum from their animals. Colostrum can be stored frozen but it does lose some of its inherent quality. Colostrum produced on a breeder's own premises is considered to be superior to colostrum from other sources, because it is produced by animals already exposed to (and, thus, making antibodies to) pathogens occurring on the premises. A German study reported that multiparous mares produced on average a liter (quart) of colostrum containing 70 grams of IgG.

    COLLECTING COLOSTRUM

    In the United States, Colostrum is taken from dairy cows with 24 hours after the birth of a calf. In most dairy cow herds, the calves are removed from their mothers soon after birth and fed colostrum from a bottle.Only dairy cows that meet USDA health standards and have been raised on a ranch with feed supplemented with nutrients can supply Colostrum. The calf needs four gallons of the nine that the cow produces. Colostrum may be collected from goats as well as cows. A USDA certified dairy collects the remaining Colostrum. The Colostrum is then frozen, taken to a processing plant, carefully thawed and evaluated for quality and immunoglobulins content. About 30-percent of the Colostrum is rejected at this point. The fat is then removed, after which the Colostrum is spray dried at low heat. The Colostrum is repeatedly tested during processing for freedom from bacterial contamination.

    Colostrum contains proteins, carbohydrates, fats, vitamins, minerals, and proteins (antibodies) that fight disease-causing agents such as bacteria and viruses. Antibody levels in colostrums can be 100 times higher than levels in regular milk. This is one of the reasons human (and other mammalian) mothers should breastfeed their newborn babies instead of relying on lab-created milk substitute formulas or other milk substitutes. The newborn(s) receives these important nutrients and antibodies from their mothers to help them survive in their environments, protecting them against disease until their own immune systems are developed enough to take over. In general, protein concentration in colostrum is substantially higher than in milk. Fat concentration is substantially higher in colostrum than in milk in some species, e.g. sheep and horses, but lower in colostrum than in milk in some other species, e.g. camels, and humans. In swine, fat concentration of milk at 48 to 72 hours after parturition may be higher than in colostrum or in late-lactation milk. Fat concentration in bovine colostrum is extremely variable.

    HUMAN CONSUMPTION OF BOVINE COLOSTRUM

    Bovine colostrum is used by humans for boosting the immune system, healing injuries, repairing nervous system damage, improving mood and sense of well being, slowing and reversing aging, and as an agent for killing bacteria and fungus. It has been used to treat diarrhea and to improve GI health. Bovine colostrum is used in the rectum to treat inflammation of the colon (colitis). People originally got interested in bovine colostrum because of the high antibody levels. They thought that the antibodies might prevent intestinal infections in people. Colostrum is collected from cows that have been vaccinated to produce antibodies that fight the bacteria that cause diarrheal disease. These antibodies appear in the colostrum that is collected as medicine. Though the hope is that these cow antibodies will help fight human disease, the cow antibodies do not seem to be very active in humans.

    Assertions that colostrum consumption is of adult human benefit are questionable because most components undergo digestion in the mature stomach, including antibodies and all other proteins. Bovine colostrum and its components are as harmful for human consumption as other bovine lactic secretions, especially in the context of intolerance or allergy to lactose or other components. Despite evidence that most components are not absorbed intact, proponents claim colostrum is useful in the treatment or prevention of a variety of illnesses.

    Colostrum contains immune factors, immunoglobulins, antibodies, proline-rich polypeptides (PRP), lactoferrin, glycoproteins, lactalbumins, cytokines (eg, interleukin, and interferon), growth factors, vitamins, and minerals. There is also a constituent isolated from bovine colostrum that is responsible for uterine and intestinal contraction, and lowering of blood pressure.

    Bovine colostrum from pasture-fed cows contains immunoglobulins specific to many human pathogens, including Escherichia coli, Cryptosporidium parvum, Shigella flexneri, Salmonella species, Staphylococcus species, and rotavirus (which causes diarrhea in infants). Before the development of antibiotics, colostrum was the main source of immunoglobulins used to fight infections. In fact, when Albert Sabin made his first oral vaccine against polio, the immunoglobulin he used came from bovine colostrum. When antibiotics began to appear, interest in colostrum waned, but, now that antibiotic-resistant strains of pathogens have developed, interest is once again returning to natural alternatives to antibiotics, namely, colostrum.

    HYPERIMMUNE BOVINE COLOSTRUM

    Researchers have created a special type of bovine colostrum called "hyperimune bovine colostrum." Hyperimmune colostrum has been an early attempt to boost the effectiveness of natural bovine colostrum by immunizing cows with a specific pathogen and then collecting the colostrum after the cow gave birth. This special colostrum is produced by cows that have received vaccinations against specific disease-causing organisms. These vaccinations cause the cows to develop antibodies to fight those specific organisms. The antibodies pass into the colostrum. Hyperimmune bovine colostrum has been used in clinical trials for treating AIDS-related diarrhea, diarrhea associated with graft versus host disease following bone marrow transplant, and rotavirus diarrhea in children.

    The U.S. Food and Drug Administration (FDA) has granted hyperimmune bovine colostrum "orphan drug status." Under the Orphan Drug Law, drug makers who invest in the development of treatments for rare conditions enjoy special market advantages; for example, permission to sell the drug without competition for 7 years. If these special incentives were not in place, pharmaceutical companies might not develop drugs for rare conditions because the potential market is so small.

    This research initially appeared very promising as antibodies did appear towards the specific pathogens or antigens that were used in the original challenge. However, upon closer examination and comparison, it was found that IgG levels in natural colostrum towards 19 specific human pathogens were just as high as in hyperimmune colostrum, and natural colostrum nearly always had higher antibody titers than did the hyperimmune version.

    IMMUNE SIGNALING PROLINE-RICH POLYPEPTIDES

    Proline-rich polypeptides are small immune signaling peptides (PRPs) were independently discovered in colostrum and other sources, such as blood plasma, in the United States, and Poland. Hence they appear under various names in the literature, including Colostrinin, CLN, transfer factor and PRP. They function as signal transducing molecules that have the unique effect of modulating the immune system, turning it up when the body comes under attack from pathogens or other disease agents, and damping it when the danger is eliminated or neutralized. At first thought to actually transfer immunity from one immune system to another, it now appears that PRPs simply stimulate cell-mediated immunity.

    IMMUNOLOGY 101- THE BASICS

    METHODS OF IMMUNE FUNCTION

    Methods of immune function can be described in two basic parts, each with several modes of action.

    1. Non-specific (innate) immunity occurs quickly and provides consistent and effective protection for the majority of pathogens. It consists of:
      A. Barriers to entry such as skin, body fluids, low pH of stomach, ciliary action of lungs, mucous linings of lungs and GI tract.
      B. Several types of Phagocytes (neutrophils, monocytes, macrophages, dendritic cells, and mast cells) that engulf (eat) non- self then kill by several methods:
      • Disinfection by hydrogen peroxide or hypochlorite.
      • Spearing the membrane electrically.
      • Breaking the cell wall with enzymes.
      • Starving them of iron.

      Phagocytes can also poison pathogens without eating them.

      A. Natural Killer cells effectively spear non- self cells like a magic bullet. They are important for destruction of pathogens.

      Note: Non-specific immune function is also important to specific immunity. Phagocytes pick up foreign cells poisoned by lymphotoxins and those identified by antibodies. Macrophages and dendritic cells present (show) antigens to plasma cells directing specific antibody production.

    When pathogens escape the non-specific defense mechanisms, specific immunity will be directed against them.

    2. The Specific (Adaptive or Acquired) immune responses.
      A. The first response of the adaptive immune system is called cell mediated. Lymphocytes are quickly turned into Killer T cells (T cells because they are regulated by the thymus). They produce substances like lymphotoxins that poison or lyse (blow up) the invader.
      B. Regulatory T suppressor cells (Ts) know when and how to shut down the killer Tís to avoid collateral damage. T helper cells (Th) help activate the 2nd response of adaptive immunity by telling B lymphocytes (think bone marrow) to produce antibody through the creation of plasma cells.
      C. The creation of antibody takes several days largely because it takes time to locate the antigen prior to creating the antibody. It is a highly specific and potent response and because of immunologic memory for both T and B cells, the second response to the same antigen is much faster and more effective than the first. This is why vaccinations are important and effective. They are used as a preventative of diseases rather than a treatment.

    CLASSES OF ANTIBODY OR IMMUNOGLOBULIN (Ig)

  • IgG antibody accounts for the vast majority of all antibody mediated protection even though it is the slowest to be produced after the initial insult. Its primary means of protection is to identify the pathogen by attaching to it and marking it for destruction by other immune cells. It can also burst cells by fixing complement. Complement is a group of proteins that build on one another after the initial component has been "fixed", much like the first domino must be tipped over to topple the whole row. Once the "row" has been completed, the complement has the ability to pierce the cell wall of the antigen. Typically only gram negative bacteria can be pierced (lysed) by a particular chain of complement known as the membrane attack (MAC). Gram positives can fall prey to the complement cascade but through the identification route.

  • IgA protects the mucosal surfaces (gut, respiratory and urogenital tract). It is similar to IgG in that it has the ability to identify and mark a pathogen for destruction by other immune cells such as phagocytes. It also has the ability to fix complement although the complement cascade is slightly different than IgG and IgM.

  • IgM is a first response antibody, offering protection from pathogens much faster than IgG upon first exposure to antigen. Like IgG it can trigger what is called the complement cascade, but is much more efficient at doing so.

  • IgE can perform the same functions as described for IgG and is effective against parasites. However it is often best known for its part in allergic reactions.

  • IgD is not as well described as the other 4 classes. It probably has a role in the activation of B cells to produce IgG by being a receptor for a broad range of antigens.

  • Note: IgE and IgD are not found in any appreciable amount in colostrum.

    Antibodies perform their mission in several ways:
    • Agglutinins cause the clumping together of bacterial cells (agglutination). This reaction has been used as a visual test for Bangís disease (sometimes called contagious abortion or undulant fever in man, Brucellosis) in cattle for many decades.
    • Bacteriolysins cause bacterial cells to dissolve or liquefy (lysis). A network of proteins known as complement assists this particular mechanism. IgG, IgA and IgE are capable of fixing complement.
    • Opsonins coat the bacteria identifying them for phagocytes.
    • Antitoxins neutralize toxins. An application of this is antiserum for snake bites.
    • Precipitins cause antigens to precipitate in a liquid media.





    COLOSTRUM USES, HEALTH BENEFITS, & SCIENTIFIC EVIDENCE

    Colostrum supplementation can boost the immune system, help burn body fat and build lean muscle. It can also speed the recovery of injuries, increase vitality and stamina. Some believe Colostrum is the ultimate anti-aging and immune supplement. It works to reduce the levels of hormones that damage the immune system and cause aging. Colostrum contains high levels of protein and is rich in immunoglobulins, which provides essential growth and immune factors, and can prevent infection.     INFECTIOUS DIARRHEA

    Infectious diarrhea. Taking bovine colostrum seems to reduce infectious diarrhea in children and people with a weakened immune system, including those with HIV/AIDS and bone marrow transplant recipients. Several studies show how bovine colostrum concentrates, including G immunoglobulin isolates, are highly successful alternative agents used to improve GI health and to treat diarrhea caused by a variety of pathogens. In AIDS patients, diarrhea and subsequent weight loss pose a problem. The severity of symptoms in some cases and sometimes unidentifiable pathogens unaffected by antibiotics welcome alternative therapy with bovine colostrum. Most clinical trials have used hyperimmune bovine colostrum. Hyperimmune bovine colostrum has FDA orphan drug status for AIDS-related diarrhea. However, some research suggests that taking hyperimmune bovine colostrum by mouth does not improve symptoms of infectious diarrhea caused by Shigella bacteria. More clinical trials need to be performed to understand and define fully the many benefits of bovine colostrums.

    COLON INFLAMMATION (COLITIS)

    There is some early evidence that a rectal enema containing 10-percent bovine colostrum might be helpful for treating colitis (inflammation of the colon). Using a specialized colostrum preparation in animal studies, bovine colostrum has been shown to have a high capacity for neutralization of bacterial toxins and high effectiveness in treating severe diarrhea. Bovine colostrum supplementation, in another report, has been shown to prevent NSAID-induced gut injury in various laboratory and animal studies, suggesting its possible usefulness for certain ulcerative bowel conditions.

    MULTIPLE SCEROSIS (MS)

    Some early evidence shows that taking hyperimmune bovine colostrum by mouth for 4 weeks does not relieve symptoms of MS. However, other early evidence shows that taking anti-measles bovine colostrum by mouth for 30 days may improve symptoms of MS.

    UPPER RESPIRATORY TRACT INFECTION

    Upper respiratory tract infection, including the common cold and flu (influenza). Early research suggests that taking bovine colostrum for 8 weeks can reduce the risk of upper respiratory tract infections in young, physically active males. Certain immune factors and antibodies also fight a variety of organisms, allergens, or toxins including pneumonia, candida, and flu. Constituent lactoferrin prevents pathogens from getting the iron they need to flourish. Lactalbumins and cytokines (interleukin and interferon) also are important as antivirals and anticancer agents.

    ATHLETIC PERFORMANCE

    The immune-boosting properties of bovine colostrum have been promoted as performance enhancers and anti-aging healing supplements. Some athletes have used bovine colostrum in an attempt to burn fat, build lean muscle, increase stamina and vitality, and improve athletic performance, decrease recovery time, and prevent sickness during peak performance levels. One clinical trial finds bovine colostrum supplement to increase serum IGF-1 concentration in athletes. IGF-1 is a growth factor that speeds up protein synthesis and slows catabolism. Developing research suggests that bovine colostrum might increase athletic performance for many, but not all, athletic activities. Activities that seem to benefit are jumping and bicycle sprints, but not running on a treadmill, rowing, or resistance training. Bovine colostrum is not on the banned drug list of the International Olympic Committee.

    LOW IGF-1 LEVELS

    Low IGF-1 levels may be associated with dementia in the very elderly, although causation has not been established. People with eating disorders also have low levels of IGF-1 due to malnutrition, as do obese individuals. Supplementation with colostrum, which is rich in IGF-1, can be a useful part of a weight reduction program. Although IGF-1 is not absorbed intact by the body, it does stimulate the production of IGF-1 when taken as a supplement.

    ANTIOXIDANT COMPONENTS

    Colostrum also has antioxidant components, such as lactoferrin and hemopexin, which binds free heme in the body.





    COLOSTRUM DOSAGE INFORMATION

    Bovine Colostrum is available in supplement form, but it is important to ask a knowledgeable source about the individual brands of Colostrum. Bovine colostrum is a difficult preparation to standardize because its antibody content may vary widely. Some clinical studies have been performed with hyperimmune colostrum, which may have a specific antibody titer; however, most products do not meet this criterion. Studies administering 25 to 125 mL of liquid formulations or 10 to 20 grams of dry powder have been reported.

    The recommended dose for adults with disease symptoms is 1,000 to 2,000 mg of the powdered Colostrum in capsules, taken twice daily with 8 to 12 ounces of water. For infections diarrhea, an oral dosage (by mouth) 10 to 20 grams of colostrum daily for 10 days has been studied in scientific research.

    Supplementation with bovine colostrum, 20 grams per day (g/d), in combination with exercise training for 8 weeks may increase bone-free lean body mass in active men and women.

    Preventive doses are left to the patientís choice.

    Children can be given Colostrum but requires less than adults.

    COLOSTRUM SAFETY & INTERACTION INFORMATION

    There are no known safety issues associated with using Colostrum supplements. There are no known interactions associated with bovine colostrum.

    Bovine colostrum appears to be safe for most people when taken appropriately by mouth and is well tolerated. When it is given rectally as an enema it seems to be possibly safe for most people. While most people do not experience any side effects from bovine colostrum, there have been rare reports of problems in HIV-positive people such as nausea, vomiting, abnormal liver function tests, and decreased red blood cells.

    There is some concern about the possibility of catching "mad cow disease" (bovine spongiform encephalitis, BSE) or other diseases from products that come from animals. "Mad cow disease" does not appear to be transmitted through milk products, but it is probably wise to avoid animal products from countries where "mad cow disease" has been found.

    Those individuals allergic to cow's milk or milk products, you may also be allergic to bovine colostrum. In that case, it is best to avoid using it to prevent an allergic reaction.

    Pregnancy and breast-feeding: There is not enough reliable information about the safety of taking bovine colostrum if you are pregnant or breast-feeding. Stay on the safe side and avoid use.
