    TMG ORGANIC COMPOUND

    Trimethylglycine (TMG) is an organic compound that occurs in plants. Trimethylglycine was the first betaine discovered by science; originally it was simply called betaine (anhydrous betaine) because, in the 19th century, it was discovered in sugar beets. Since then, many other betaines have been discovered, and the more specific name glycine betaine distinguishes this one.

    STRUCTURE & REACTIONS

    Trimethylglycine is an N-trimethylated amino acid. This quaternary ammonium exists as the zwitterion at neutral pH. Strong acids such as hydrochloric acid converts TMG to the salt betaine hydrochloride:
      (CH3)3N+CH2CO2- + HCl —> [(CH3)3N+CH2CO2H]Cl-
      Demethylation of TMG gives dimethylglycine. Degradation of TMG yields trimethylamine, the scent of putrifying fish.

    PRODUCTION & BIOCHEMICAL PROCESSES

    Processing sucrose from sugar beets yields glycine betaine as a byproduct. The value of the TMG rivals that of the sugar content in sugar beets. Glycine betaine production involves chromatographic separation. In most organisms, glycine betaine is biosynthesized by oxidation of choline in two steps. The intermediate, betaine aldehyde, is generated by the action of the enzyme mitochondrial choline oxidase (choline dehydrogenase, EC 1.1.99.1). Betaine aldehyde is further oxidised in the mitochondria or cytoplasm to betaine by the enzyme called betaine aldehyde dehydrogenase (EC 1.1.1.8).

    TMG is an organic osmolyte that occurs in high concentrations (10s of millimolar) in many marine invertebrates, such as crustaceans and molluscs. It serves as a potent appetitive attractant to generalist carnivores such as the predatory sea-slug Pleurobranchaea californica.

    TMG is an important cofactor in methylation, a process that occurs in every cell of mammals to synthesize and donate methyl groups (CH3) for other processes in the body. These processes include the synthesis of neurotransmitters such as dopamine, serotonin. Methylation is also required for the biosynthesis of melatonin and the electron transport chain constituent coenzyme Q10. The major step in the methylation cycle is the remethylation of homocysteine, which can occur via either of two pathways. The major pathway involves the enzyme methionine synthase, which requires vitamin B-12 as a cofactor, and also depends indirectly on folate and various other B vitamins. The minor pathway involves betaine-homocysteine methyltransferase and requires TMG as a cofactor. Betaine is thus involved in the synthesis of many biologically important molecules, and may be even more important in situations where the major pathway for the regeneration of methionine from homocysteine has been compromised by genetic polymorphisms.

    TMG IN AGRICULTURE & AQUACULTURE

    Factory farms supplement fodder with TMG and lysine to increase livestocks' muscle mass (and, therefore, "carcass yield", the amount of usable meat). Salmon farms apply TMG to relieve the osmotic pressure on salmons' cells when workers transfer the fish from freshwater to saltwater. TMG supplementation decreases the amount of adipose tissue in pigs; however, research in human subjects has shown no effect on body weight, body composition, or resting energy expenditure.

    TRIMETHYLGLYCINE RESEARCH

    Trimethylglycine (Betaine) supplementation decreases plasma homocysteine concentrations but does not affect body weight, body composition, or resting energy expenditure in human subjects. (Am J Clin Nutr. 2002.)

    Betaine (trimethylglycine) is found in several tissues in humans. It is involved in homocysteine metabolism as an alternative methyl donor and is used in the treatment of homocystinuria in humans. In pigs, trimethylglycine decreases the amount of adipose tissue. The aim of the study was to examine the effect of trimethylglycine supplementation on body weight, body composition, plasma homocysteine concentrations, blood pressure, and serum total and lipoprotein lipids. Forty-two obese, white subjects (14 men, 28 women) treated with a hypoenergetic diet were randomly assigned to a trimethylglycine-supplemented group (6 g/d) or a control group given placebo for 12 weeks. The intervention period was preceded by a 4-week run-in period with a euenergetic diet. Body weight, resting energy expenditure, and fat mass decreased significantly in both groups with no significant difference between the groups. Plasma homocysteine concentrations decreased in the trimethylglycine group. Diastolic blood pressure decreased without a significant difference between the groups. Serum total and LDL-cholesterol concentrations were higher in the trimethylglycine group than in the control group. A hypoenergetic diet with trimethylglycine supplementation (6 grams daily for 12 weeks) decreased the plasma homocysteine concentration but did not affect body composition more than a hypoenergetic diet without trimethylglycine supplementation did.

    TMG supplementation may cause diarrhea, stomach upset, or nausea. Obese persons or those with kidney disease supplementing with TMG, folic acid, and vitamin B-6 can experience an increase in total cholesterol levels.

    DMG VS TMG INFORMATION

    DMG (dimethylglycine) is basically the amino acid glycine attached to two methyl groups while TMG, also known as betaine, has three methyl groups. Commonly these methyl groups are added to homocysteine, converting it into the amino acid methionine which is then converted to SAMe, a nutrient available over the counter which has been used as a powerful antidepressant. DMG plays a crucial role in the respiratory cycle of the cells, transporting oxygen and serving as a potent methyl donor. DMG acts as a building block for the synthesis of many important substances such as choline, SAM-e, the amino acid methionine, several hormones, neurotransmitters, and DNA and is more beneficial for children with autism. The formation of the neurotransmitters norepinephrine and dopamine requires a methyl group donated by SAMe. Dopamine enhances sex drive. DMG is very similar to TMG, but it is not a methyl-dontain agent like TMG and are involved in different bioactive reactions. Therefore DMG should not be considered a substitute for TMG.

    DIMETHYLGLYCINE (DMG)

    Dimethylglycine (DMG) is a derivative of glycine, the simplest of the amino acids. It is a non-protein amino acid found naturally in animal and plant cells. DMG is produced in cells as an intermediate in the metabolism of choline to glycine. It acts as a building block for many important substances, including the amino acid methionine, choline, a number of important hormones and neurotransmitters, and DNA.

    There has been much confusion surrounding the history of DMG as a nutritional supplement. DMG appeared as a supplement in the 1960s under the names vitamin B-15, pangamic acid and calcium pangamate. Calcium pangamate was originally a mixture of calcium gluconate and DMG. Calcium pangamate was intended as a delivery form of DMG. However, several products entered the supplement marketplace called pangamic acid or calcium pangamate, and these did not contain DMG. Some of these products contained, instead of DMG, a substance called diisopropylammonium dichloroacetate. At present, DMG supplements are available that do contain dimethylglycine.

    DMG-containing calcium pangamate was popular with Russian athletes and cosmonauts because it was reputed to enhance oxygenation at the cellular level, reduce fatigue and enhance physical stamina. None of those claims, however, was ever substantiated. DMG is neither a vitamin nor an essential nutrient. DMG is also known as N, N-dimethylglycine, (dimethylamino) acetic acid and N-methylsarcosine.

    DMG is a solid, water-soluble substance. DMG should not be confused with TMG (trimethylglycine or betaine). TMG is involved in the methylation of homocysteine to form methionine (See Trimethylglycine). DMG can be formed from TMG upon the loss of one of its methyl groups.

    DMG is absorbed from the small intestine and from there transported by the portal circulation to the liver. DMG is swiftly metabolized in the liver converting dimethylglycine into other useful metabolites by means of oxidative dimethylation. DMG metabolizes to monomethylglycine or sarcosine which, in turn, is converted to glycine. Dimethylglycine dehydrogenase, a flavoprotein, is the enzyme that catalyzes the oxidative demethylation of DMG to sarcosine. The methyl group produced in this reaction returns to the one carbon pool at the level of N10-hydroxymethyl-tetrahydrofolic acid. DMG itself is formed from trimethylglycine or betaine. DMG that is not metabolized in the liver is transported by the circulatory system to various tissues in the body. There are no known deficiency symptoms due to a lack of DMG in the diet; however, taking this amino acid does offer a wide range of health benefits.

    DMG serves as a building block to amino acids, hormones, neurotransmitters, and even DNA. Taking supplemental DMG can have a wide range of possible beneficial effects. DMG has been suggested for use as an athletic performance enhancer, immunostimulant, and a treatment for autism, epilepsy, or mitochondrial disease. It is thought to be a metabolic enhancer and aid in processes associated with those included in cellular respiration, immune response, and oxygen utilization within the body. It helps the body maintain high energy levels and boost mental acuity. DMG has been found to enhance the immune system and to reduce elevated blood cholesterol and triglyceride levels. It improves oxygen utilization by the body, helps to normalize the functioning of many important organs. Dimethylglycing may also play equally important roles in the prevention of fatigue and improvements in liver function. It may also be useful for controlling epileptic seizures.

    Trimethylglycine (TMG) is found in a variety of plant and animal sources and is utilized in the conversion of homocysteine to methionine. Maintaining normal homocysteine levels is important for the health of the cardiovascular system. TMG helps convert homocysteine to methionine by donating a methyl (CH3) group.

    Reducing Homocysteine Levels with Trimethylglycine: Elevated homocysteine levels are easily reduced with the intake of certain nutrients involved in methylation. Such nutrients include folic acid with its cofactors, vitamins B-6 and B-12, plus what is proving to be the most efficient and cost-effective methyl donor of all, Trimethylglycine (TMG).

    TMG (also known as anhydrous betaine) is a naturally occurring phytonutrient found in a variety of plants and animals, with highest concentrations in beets, leafy green vegetables, and legumes. Since it can be difficult to eat enough of these foods to provide the body with sufficient methyl groups, supplementation may be necessary.     METHYLATION & BENEFITS

    A methyl donor is simply any substance that can transfer a methyl group [a carbon atom attached to three hydrogen atoms (CH3)] to another substance. Many important biochemical processes rely on methylation, including the metabolism of lipids and DNA. Scientists suspect that adequate methylation of DNA can prevent the expression of harmful genes, such as cancer genes. It is quite likely that our body's ability to methylate declines with age, potentially contributing to the aging process, and therefore supplementation is an interesting idea in terms of health benefits.

    Methylation is a normal biological process that occurs in healthy human bodies every minute of every day. Methylation is an important step in the conversion of nomocysteine, a damaging amino acid, into the biologically safe and beneficial amino acid, methionine. In addition, methylation is required for DNA repair, as well as for the production of SAMe, which helps maintain healthy joint tissue. Methylation can be inhibited by poor or inadequate diet and unhealthy lifestyle practices. There are no deficiency symptoms known. TMG (Trimethylglycine) has been shown to be a safe and effective methyl donor for the facilitation of methylation processes. TMG supplementation has been shown in clinical and non-clinical studies to support a healthy homocysteine level, which in turn supports healthy cardiovascular function. Elevated homocystein levels increase risk factors for cardiovascular disease, heart attack, stroke, neural tube defects, cancer, liver disease, depression and peripheral neuropathy.

    Although TMG supplementation decreases the amount of adipose tissue in pigs, research on human subjects has shown no effect on body weight, body composition, or resting energy expenditure when used in conjunction with a hypoenergetic diet. The Food and Drug Administration of the United States approved anhydrous trimethylglycine (also known by the brand name Cystadane) for the treatment of homocystinuria, a disease caused by abnormally high homocysteine levels at birth. TMG lowers homocyteine levels.

    ROLE FOR ANTI-AGING

    Trimethylglycine is a methyl donor supplement that assists in methylation and may help protect cellular DNA from mutation. Trimethylglycine helps to keep the liver healthy by assisting the detoxification process, and animal studies suggest that the compound's methyl group-donating properties may help to protect the liver cells from toxins and chemical damage. TMG is a safe and well tolerated nutrient that leads to a significan biochemical and historical improvement in people with non-alcoholic steatohepatitis (NASH). Laboratory studies and two clinical trials have indicated that TMG is a potential treatment of non-alcoholic steatohepatitis. TMG may help protect the liver agains the effects of alcohol, perhaps by stimulating the formation of SAM-e. In addition it may be helpful for non-alcoholic forms of fatty liver (non-alcoholic steatosis) as well.

    TMG is sometimes used as a treatment for depression, as it can increase S-adenosylmethionine (SAMe) by remethylating homocysteine. SAMe has been shown to work as a nonspecific antidepressant in several studies.

    Trimethylglycine aids in the conversion of homocysteine to methionine, and thus helps to lower levels of the amino acid homocysteine, high levels of which have been linked to an increased risk of heart disease, heart attack, stroke, Alzheimerís disease, Parkinsonís disease, and osteoporosis. At this time the clinical uses of Trimethylglycine and DMG are not well defined and whether they would be helpful in the therapy of Alzheimer and Parkinsonís disease is not known. Since the bodyís ability to methylate declines with age, supplements in small amounts, such as 50 to 100 mg a day, may benefit middle aged and older individuals. It is recommended that a dosage should not exceed 250 mg of Trimethylglycine or DMG on a daily basis until more is known about these supplements. Your dose of trimethylglycine and DMG should be reduced if you are taking B vitamins, SAMe, DMAE, or choline since all of these nutrients have overlapping functions.

    SCIENTIFIC USES

    PCR - Trimethylglycine can act as an adjuvant of the polymerase chain reaction (PCR) process, and other DNA polymerase-based assays such as DNA sequencing. By an unknown mechanism, it aids in the prevention of secondary structures in the DNA molecules, and prevents problems associated with the amplification and sequencing of GC-rich regions. Trimethylglycine makes guanosine and cytidine (strong binders) behave with thermodynamics similar to those of thymidine and adenosine (weak binders). It has been determined under experiment that it is best used at a final concentration of 1M.

    IEX Ion Exchange Chromatography - In the book from Amersham Biosciences/GE Healthcare, Ion Exchange Chromatography & Chromatofocusing - Principles and Methods, page 48 - Zwitterionic additives such as betaine can prevent precipitation and can be used at high concentrations without interfering with the gradient elution.

    SOURCES

    TMG is a natural compound found in small quantities in some plant foods. However, humans do not need to obtain it from their diet because the body can manufacture it from other nutrients. The best known natural sources of TMG are plants from the chenopodiaceae family (the sugar beet). A diet high in broccoli, spinach or beets can provide as much as a fifth or even a quarter teaspoon of TMG, just over 500 mg. Grains, nuts, seeds and meats contain small amounts of TMG. However, most TMG in food is destroyed during cooking or processing, so food is not usually a reliable way to get therapeutic dosages. In its natural form, TMG is a white crystal with a distinctive mildly sweet taste and aftertaste. It may be dissolved into juice, water or tea.

    SUPPLEMENT FORMS

    No optimal therapeutic dosage of TMG has been established.

    Product manufacturers recommend dosages between 375 and 1,000 mg daily up to 3,000 mg daily. Refer to product packaging for use directions. A maximum safe level has not been established. Note: TMG is very hydroscopic. This means it absorbs water from the atmosphere and tends to split capsules from expansion which is why many companies tablet it. Keep this in mind when buying, using and storing TMG supplements.



    SOME SUGGESTED DOSAGES FOR CERTAIN HEALTH CONDITIONS

  • Anti-Depression: By raising level of beneficial SAM (S-Adenosyl-Methionine). Dose range: 500 to 1,500 mg daily.

  • Body Building: In the animal husbandry field, TMG is used to decrease fat and increase meat yield. While human studies have just started, a 200 pound individual with 20 percent body fat can expect to lose as much as 5 pounds of fat and gain as much as 12 pounds of muscle. Dose range: 500 to 2,000 mg daily.

  • Cardiac Protection: Conversion of homocysteine to Methionine. Dose range: 500 to 1,000 mg daily.

  • General Health Preventative: As part of a formula for maintaining good health or disease prevention. Dose range: 500 to 1,000 mg daily.

  • Glutathione Elevation: In several different studies, TMG has been shown to increase hepatic Glutathione, the body's most important antioxidant. Dose range: 500 to 1,500 mg daily.

  • Homocysteine Lowering: Specific for that purpose. The supplement of choice for lowering homocystenuria, Dose range: 500 to 1,000 mg daily. As used by clinicians and hospitals with patient monitoring -up to 3 grams daily.

  • Liver Disorders: As a part of liver-healing and protection formulas. Increases SAM levels in the liver, enables the liver to metabolize fat and protect against many challenges such as alcohol induced cirrhosis. TMG will also decrease bilirubin, alkaline phosphotase, and several other liver enzymes related to a large variety of liver disorders. Significant liver benefits have been shown in 20 studies. Dose range: 500 to 1,500 mg. daily.

    Frequently dosing of supplements are based on adults. As we all know, children have smaller body size and thus require different dosage of many vitamins and supplements, especially medications. Here is a formula that you can use simply to estimate the dose for children:
      Adult dose divided by 150, then multiply by your child's weight in pounds. This will give you the dose for your child.

    SAFETY CONCERNS

    No contraindications known.

    Do not confuse Betaine TMG with Betaine HCl. TMG is similar chemically to Betaine Hydrochloride, but it has entirely different actions. Some supplement companies have mistakenly labeled their products. If found in capsules, open the capsule and taste the ingredients. If it is strongly acidic, it is most likely Betaine HCl or hydrochloric acid. Similarly, chewing a tablet should not burn your tongue but have a mildly sweet taste.

    TMG taken in high dosages, such as more than 750 mg, can cause nausea, increased body temperature, irritability, restlessness and insomnia and perhaps muscle tension headaches. People with kidney disease or liver disease, young children and pregnant or breastfeeding women should consult with their health care provider before taking TMG. Maximum safe dosages have nto been established for these groups of individuals. People with high or borderline-high cholesterol should use TMG only with caution.

    It may be recommended in conjunction with Vitamin B-12 injections for cognitive abilities in children. It is easy for children to overdose on TMG and should only be used by children under the supervision of a health care provider.

    When using TMG supplements, each person is unique in their nutritional requirements and response to supplement use. Some require more, others less.
